You know what, I always thought if this was the early 90's, I totally think the actor that played the T-1000 in Terminator 2: Judgement Day would be closest to an ideal live-action version of Wesker (hell, he had the shades and the cold, creepy stare down). I've always thought of this even before the first Resident Evil movie came out.
